Molecular structure

Modified GRF (1-29), commonly sold as CJC-1295 without DAC, is a 29-residue peptide with four substitutions relative to the corresponding native GHRH fragment: D-Ala at position 2, Gln at position 8, Ala at position 15, and Leu at position 27. It has a C-terminal amide.

Ipamorelin is a modified pentapeptide commonly represented as Aib-His-D-2-Nal-D-Phe-Lys-NH₂. Its nonstandard residues and stereochemistry are integral to its chemical identity.

Physicochemical properties

The two peptides differ substantially in molecular size and sequence. Their solubility, charge, and stability are therefore not identical. The behavior of the combined preparation depends on the formulation conditions and the chemical forms of both constituents.

Biochemical mechanisms

Modified GRF (1-29) interacts with the GHRH receptor, while ipamorelin is associated with the ghrelin receptor, also called GHS-R1a. These are distinct G-protein-coupled receptors.

The two peptides therefore have different primary receptor targets, even though their signaling can converge on downstream cellular processes.
