Molecular structure

Tirzepatide is a synthetic 39-residue peptide containing nonstandard amino-acid modifications and a fatty-diacid component connected to the peptide through a linker. The peptide has a C-terminal amide and a defined sequence related to incretin peptides.

The fatty-diacid component is covalently connected to a lysine side chain. This modification changes the molecule's hydrophobic characteristics and its capacity to interact with proteins such as albumin.

The peptide backbone provides the principal structural features responsible for receptor recognition, while the lipid-related modification alters the molecule's physicochemical and binding properties.

Physicochemical properties

The molecule combines a highly polar peptide backbone with a comparatively hydrophobic lipid-related group. Its overall behavior depends on the distribution of charged residues, the linker, and the attached fatty-diacid structure.

The covalent modification is part of the molecule itself, not simply an ingredient mixed with the peptide. Removing or altering it would produce a chemically different structure.

Biochemical mechanisms

Tirzepatide activates the GIP receptor and GLP-1 receptor. Both belong to the glucagon receptor family of G-protein-coupled receptors and can signal through Gs proteins and adenylyl cyclase, increasing intracellular cyclic AMP.

The molecule's dual-receptor activity distinguishes it from retatrutide, which is designed to interact with three receptor types.

Scientific evidence

The molecular formula, molecular weight, peptide sequence, and dual-receptor classification are established features. The identity of an individual sample still depends on confirmation of the complete modified structure.

GLP2-T is supplied strictly for research use only and is not intended for human or animal consumption, clinical application, diagnostic use, or therapeutic purposes.
